No, since Sass has been shown that Usagi is Moon, it probably wasn't
hard for her to come to the conclusion that Makoto is probably a
Senshi. I think she even speculated about it on-screen. And even
if Makoto isn't a Senshi, she's still connected to Usagi, and as
such Yoshi represents leverage, even if once- or twice-removed.
Sass doubtless has APBs out on all the senshi, all the civilian IDs
she's been able to connect to them, and everyone she suspects is
connected to them at all. Yoshi just came up onto her radar first.
All the Sisterhood as a whole has on Yoshi is that he's a bystander
who poked his nose where it didn't belong. Their resources are
finite so they have to limit the search at some point. Vi, on the
other hand, has a special bone to pick with him.
I think it was made very clear during the confrontation that Yoshi is not
just a bystander who happened upon a bad situation. Check out this excerpt
from chapter 7:
"I don't think you should be doing that," a curiously gentle
voice said, and Makoto turned her head to see him standing there,
dressed in blue jeans and a white t-shirt.
Yoshi.
"Well, well," the woman snarled. "What have we here, a
knight in shining armour? This doesn't concern you, boy. You
should leave while you have the chance."
"If it concerns her, it concerns me," Yoshi replied, his
stance loose and deceptively relaxed. Makoto wanted to warn him,
but as she got some of her wind back, she realized that the Sister's
attention was mostly focussed on him.
And later:
"Look out!" Yoshi cried, and suddenly Makoto found
herself thrown to the side as the werewolf threw her out of the way.
She rolled across the pavement, grateful for the protection afforded
by the leather she wore. As it was, she picked up more bruises as
she skidded across the rough parking lot. When she tumbled to a
stop, she managed to rise to all fours, lifting her head to see Yoshi
standing where she'd been, a dazed expression on his face. Slowly
he reached up and pulled a black rose from the base of his throat
where it had lodged, stem first, and clumsily let it fall from a limp
hand.
And:
"Well, that wasn't what I had in mind," she said softly, her
eyes pinning Makoto with malicious glee. "But this will work out
well, I think. Yes, I'll just have your boyfriend bring you to heel
for me. Nobody can resist the enchantment of our Black Rose. He
will now obey me, do anything to please me. And I do mean
anything ..."
"If it concerns her, it concerns me." And he throws himself between Makoto
and Vi's attack. Makoto calls him by name, and Vi refers to him as Makoto's
boyfriend. It's clear that to her that this is no random bystander, but
someone who cares enough for Makoto to risk his life for her. That makes him
good bait at the very least, a potential source of valuable info if they're
lucky.
